根據(jù)上述培養(yǎng)基配方,配好每一種常量營養(yǎng)鹽類1000倍的濃縮儲備液。同時(shí)配制微量營養(yǎng)混合鹽類1000倍的濃縮儲備液TMS(Trace?Metal?Solution)。在900ml蒸留水中依次加入各儲備液1ml,添加順序?yàn)椋篘aNO3(10ml)→KNO3→K2HPO4→MgSO4·7H2O→Na2CO3→Ca(NO3)2·4H2O→NH4Cl(NH4Cl-?EDTA)→Fe-EDTA??→TMS。每加入一種營養(yǎng)鹽必須充分搖動(dòng)容量瓶,使混合均勻后再加入下一種。
