經(jīng)修改的HGZ-145培養(yǎng)基配方用于雨生紅球藻培養(yǎng)。

**:?NH4Cl(NH4Cl-?EDTA)與Fe-EDTA配置的方法參照文獻(xiàn)葉居新等(1999)。

根據(jù)上述培養(yǎng)基配方,配好每一種常量營(yíng)養(yǎng)鹽類(lèi)1000倍的濃縮儲(chǔ)備液。同時(shí)配制微量營(yíng)養(yǎng)混合鹽類(lèi)1000倍的濃縮儲(chǔ)備液TMSTrace?Metal?Solution)。在900ml蒸留水中依次加入各儲(chǔ)備液1ml,添加順序?yàn)椋篘aNO3(10ml)KNO3→K2HPO4→MgSO4·7H2ONa2CO3→Ca(NO3)2·4H2O→NH4Cl(NH4Cl-?EDTA)→Fe-EDTA??→TMS。每加入一種營(yíng)養(yǎng)鹽必須充分搖動(dòng)容量瓶,使混合均勻后再加入下一種。

1. Begin stirring and heating approximately 950 mL of dH2O.
2. When the stirring water approaches the boiling point, add the following components in the order listed.
3. Bring the volume to 1 L with dH2O.
4. Store at refrigerator temperature.
